Scientists in almost any field of biology agree that antibodies are the rate-limiting step in basic and clinical research, but the use of antibodies as human therapeutics is growing. For instance, there were 18 monoclonal-based compounds recently approved by the Food and Drug Administration (FDA) and more than 150 are currently under development. There is much interest in finding new and effective ways to develop high-affinity antibodies for both basic research and clinical applications. For instance, Stephen Johnston, head of the Center for Innovations in Medicine at Arizona State University, was pleasantly surprised to find that he and his colleagues could avoid the time-consuming aspects of large-scale protein production by simply inoculating animals with DNA encoding a protein of interest. Many researchers and some companies use the technique, including GENOVAC. Among topics covered are the work of a team at Mirus Bio on genetic immunization based on an existing gene therapy approach, molecular approaches, Cambridge Antibody Technology's recommendation of use of all natural libraries, use of CDR grafting to generate humanized antibodies, and platforms such as the 8200 Cellular Detection System from Applied Biosystems (which is an example of a type of system that automates and speeds hybridoma and monoclonal screening).
