Investigators with Erasmus University Medical Center in Rotterdam, the Netherlands, recently reported in Analytical Chemistry on a study in which they explored the potential of Raman spectroscopy for distinguishing between meningiomas and healthy dura tissue. An in vivo Raman-based method could offer real-time guidance during surgical resection of the tumors and reduce the change of recurrence. Raman methods are not invasive and do not require extrinsic contrast agents. They are therefore readily adaptable to in vivo use, and several groups have made considerable progress in developing such methods for in vivo applications using fiber-optic probes. Topics covered include distinguishing dura and challenges to be overcome. The researchers did 38 Raman experiments on samples from 20 patients. They compared the resulting pseudocolor maps with the histopathological evaluation of the same samples to correlate the spectral features seen with their histological origins. They developed a classification model to distinguish between dura and meningioma with a 100% accuracy rate during the in vitro study. Challenges are described that are related to differences in experimental condition and the need to detect tumor tissue when it is only a small fraction of the tissue volume from which a spectrum is record.
