Raman spectroscopy is now automated so that it can be used for the speedy discovery of new drugs. Thanks to recent developments in Raman lasers, beam path optics, gratings, Rayleigh rejection filters, CCD detectors, and automated alignment and calibration routines, Raman is useful not only in the research laboratory, but in routine analysis environments also. Users also can now preprocess data from sample arrays to do cluster analysis that groups like compounds and identifies possibly new compound forms. They can also present results in a format that is visualized without difficulty for rapid comprehension and assessment. Analysis of the results from a parallel experimental format requires a record of the result from each well and also a cross-correlation and overall analysis of data from all wells. Raman spectroscopy is being considered by many as another useful technique for polymorph analysis. Raman spectroscopys advantages for polymorph screening include delivery of high information content, the ability to provide molecular identification, and high sensitivity to crystalline geometry. Minimal or no sample preparation is required, which accelerates analysis because handling is reduced, and crystals are maintained in their initial form. A Raman microscope with a motorized stage is a good choice for automated data collection for high-throughput crystallization screening studies, assuming that the software provides flexible data collection methods and can support various sample array formats.
