Researchers at Ohio State University in Columbus, Ohio, have demonstrated that laser scanning cytometry can quantify chemotaxis. It can count cells, but can also provide convenient and advanced in situ analysis of cellular parameters on migrated, undisturbed cells. It uses both the statistical power of flow cytometry and accuracy of in situ fluorescence microscopy. While manual counting of cells on a membrane is tedious and error-prone, an automated system can detach cells from membranes loses information on cell positioning and morphology. Using a laser scanning cytometry instrument from CompuCyte to analyze chemotaxis results, investigators could collect bulk information on the whole population and could retain the position of each cell and its fluorescence parameters on three channels. CompuCyte explains that laser scanning cytometry is different from flow cytometry because the former works with specimens on a solid substrate. Because the depth of field is between 20-30 mu m, there is no need to refocus when large sample areas are scanned. Investigators manually counted five fields of view, directly from the filter and working from photomicrographs. The latter technique is probably the more accurate of the two.
